The survivin mRNA vaccine is an experimental therapeutic cancer vaccine designed to treat multiple myeloma (MM) by targeting the tumor-associated antigen survivin (BIRC5). The specific formulation described in recent research utilizes the "Galsome" platform, which consists of lipid nanoparticles (LNPs) containing mRNA encoding survivin and the adjuvant alpha-galactosylceramide (α-GalCer). This dual-action approach is intended to activate both survivin-specific CD8+ T cells and invariant natural killer T (iNKT) cells via the CD1d pathway. While preclinical studies in mouse models have shown a reduction in MM cell levels in the bone marrow and spleen, researchers have observed that survivin expression in activated T cells may lead to unintended reduction of the effector cell populations following boost vaccinations, suggesting a need for careful antigen selection or combination therapies.
