Survivin UsiRNA-DiLA2 is an experimental RNA interference (RNAi) therapeutic developed by MDRNA Inc. (later Marina Biotech). The drug consists of a "UsiRNA"—a small interfering RNA (siRNA) construct incorporating unlocked nucleobase analogs (UNA) to enhance stability and reduce off-target effects—designed to silence the expression of the survivin (BIRC5) gene. Survivin is an inhibitor of apoptosis (IAP) protein that is overexpressed in many malignancies, where it promotes cell survival and regulates mitosis. The UsiRNA is delivered using the proprietary Di-alkylated Amino Acid (DiLA2) liposomal platform, which facilitates systemic or local delivery to tumor tissues. Preclinical studies demonstrated significant knockdown of survivin mRNA and inhibition of tumor growth in models of bladder cancer and hepatocellular carcinoma.
