Sushi-IL15-ApoA-I is a recombinant triple fusion protein designed for cancer immunotherapy. It consists of human interleukin-15 (IL-15), the sushi domain of the IL-15 receptor alpha (IL-15Rα), and apolipoprotein A-I (ApoA-I). The ApoA-I component allows the molecule to integrate into circulating high-density lipoproteins (HDLs), significantly extending the plasma half-life of the IL-15 cytokine. The sushi domain of IL-15Rα facilitates the trans-presentation of IL-15 to the IL-15 receptor beta/gamma (IL-15Rβ/γ) complex, leading to the potent activation and expansion of natural killer (NK) cells and memory CD8+ T cells. Developed primarily at the Foundation for Applied Medical Research (CIMA), University of Navarra, it has shown preclinical efficacy in metastatic melanoma and colon cancer models, both as a monotherapy and in combination with other agents like cetuximab.
