Suvesitetamab (SIM0501) is a humanized IgG1 monoclonal antibody targeting Growth Differentiation Factor 15 (GDF15), a stress-induced cytokine belonging to the TGF-beta superfamily. GDF15 is frequently upregulated in various malignancies, including colorectal, lung, and prostate cancers, where it contributes to immune suppression, tumor progression, and cancer-associated cachexia by binding to its receptor, GFRAL. By neutralizing circulating GDF15, suvesitetamab aims to reverse immune evasion and metabolic dysfunction in the tumor microenvironment. Developed by Simcere Pharmaceutical, the agent is currently being investigated in clinical trials as both a monotherapy for advanced solid tumors and in combination with chemotherapy (e.g., trifluridine/tipiracil) for refractory metastatic colorectal cancer.
