SUVN-502 + donepezil + memantine is a triple combination therapy investigated for the symptomatic treatment of moderate Alzheimer's disease. The combination includes: - Masupirdine (SUVN-502), a selective 5-hydroxytryptamine 6 (5-HT6) receptor antagonist, which modulates neurotransmitter release and aims to enhance cognitive function. - Donepezil, an acetylcholinesterase inhibitor that increases acetylcholine levels in the brain to improve cognition. - Memantine, an N-methyl-D-aspartate (NMDA) receptor antagonist that regulates glutamatergic activity to protect against excitotoxicity. This triple therapy was evaluated in a phase 2a proof-of-concept study as adjunctive treatment in patients with moderate Alzheimer's disease already receiving stable doses of donepezil and memantine. While the primary endpoint was not met, subgroup analyses suggested potential benefits on cognition and behavioral symptoms in certain patient populations. The regimen was generally safe and well tolerated[1][3][4][5].
