Overview

SUVN-I6107 is a novel, potent, and selective small molecule muscarinic M1 receptor positive allosteric modulator (M1 PAM) with minimal agonist-like activity. It is being developed by Suven Life Sciences for the treatment of cognitive disorders, primarily Alzheimer's disease. Unlike direct agonists or cholinesterase inhibitors, SUVN-I6107 enhances the activity of endogenous acetylcholine at the M1 receptor without significant affinity for other muscarinic subtypes (M2–M5), aiming to improve cognition while minimizing cholinergic side effects such as salivation or diarrhea. Preclinical studies have demonstrated robust efficacy in animal models of cognition and favorable safety margins. The drug exhibits excellent pharmacokinetic properties including good brain penetration and high cerebrospinal fluid concentrations in nonclinical species. As of 2025, it is undergoing Phase 1 clinical trials to assess its safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy subjects[1][2][4][5][6][8].
