Overview

sv-SLC12A7 CAR-T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target a leukemia-specific splice variant of the solute carrier family 12 member 7 (SLC12A7) protein. Developed by researchers at the H. Lee Moffitt Cancer Center and collaborating institutions, this therapy utilizes a 4-1BB/CD3z (BBz) costimulatory architecture. The target, sv-SLC12A7, is an isoform generated by alternative mRNA splicing that is highly expressed on the surface of acute myeloid leukemia (AML) blasts and leukemic stem cells (LSCs), while remaining absent or minimally expressed in normal hematopoietic stem cells and non-hematopoietic tissues. In preclinical xenograft mouse models, these CAR-T cells demonstrated effective eradication of THP1 leukemic cells, suggesting a potential strategy to minimize on-target off-tumor toxicity in AML treatment.
