SV201 is a preclinical-stage bispecific nanobody candidate developed by Sysvax for the treatment of HER2-positive breast cancer. As a nanobody-based therapeutic, it utilizes single-domain antibody fragments (VHH) typically derived from camelids, which are significantly smaller than conventional antibodies, allowing for potentially better tumor penetration and the ability to bind to cryptic epitopes. SV201 is engineered to be bispecific, meaning it targets two distinct epitopes on the Human Epidermal Growth Factor Receptor 2 (HER2) simultaneously. This biparatopic binding strategy is designed to enhance the inhibition of HER2-mediated signaling pathways, promote receptor internalization and degradation, and potentially overcome resistance mechanisms associated with standard-of-care monospecific antibodies like trastuzumab.
