SVC-KRAb is a first-in-class live biotherapeutic cancer therapy developed by SiVec Biotechnologies that targets oncogenic RAS mutations. The drug utilizes the company's proprietary BactPac platform, which employs engineered non-pathogenic bacteria to deliver circular messenger RNA (mRNA) directly into tumor cells. Once inside the tumor environment, the circular mRNA is translated to produce a nanobody (single-domain antibody) that targets and inhibits over 20 cancer-driving mutant RAS proteins, including pan-RAS (KRAS, HRAS, NRAS) and specific mutations like KRAS G12S and KRAS G12V. SVC-KRAb is designed to treat KRAS-driven malignancies such as pancreatic ductal adenocarcinoma (PDAC), colorectal cancer (CRC), and non-small cell lung cancer (NSCLC). In preclinical studies, the therapy demonstrated significant tumor volume reduction in xenograft models and is currently undergoing IND-enabling studies.
