SVC112 is a synthetic small molecule derivative of bouvardin, a natural compound found in the firecracker bush (Bouvardia ternifolia). It functions as a translation elongation inhibitor, specifically targeting protein synthesis in cancer stem cells (CSCs) while sparing healthy cells. SVC112 has demonstrated potent antiproliferative effects against head and neck squamous cell carcinoma (HNSCC), colorectal cancer, melanoma, non-small cell lung cancer, salivary gland cancer, and leukemia models. Mechanistically, it inhibits ribosomal processing of mRNAs critical for stress response and reduces CSC-related proteins such as Myc and Sox2. Preclinical studies show that SVC112 enhances the effects of radiation therapy by depleting CSCs and delaying DNA repair in tumor cells. Compared to FDA-approved protein synthesis inhibitors like omacetaxine mepesuccinate (HHT), SVC112 exhibits greater selectivity for malignant cells with reduced toxicity to normal tissues[4][5][6].
