Overview

SVV-012 is an armed oncolytic virus developed by Seneca Therapeutics for the treatment of solid cancers. It is an engineered version of the Seneca Valley Virus (SVV-001), a non-pathogenic picornavirus that naturally exhibits tropism for Tumor Endothelial Marker 8 (TEM8), also known as Anthrax Toxin Receptor 1 (ANTXR1). TEM8 is highly expressed on the surface of various cancer cells and tumor-associated endothelial cells but has limited expression in normal tissues, allowing for selective viral infection and replication. As an "armed" virus, SVV-012 is modified to carry therapeutic transgenes, such as immune-stimulating cytokines or other modulators, which are expressed locally within the tumor microenvironment upon infection. This dual mechanism of action—direct oncolysis and transgene-mediated immune activation—is intended to overcome the immunosuppressive nature of solid tumors. It is currently in preclinical development.
