Overview

SW-682 is an investigational, orally bioavailable small molecule inhibitor of the TEA domain (TEAD) family of transcription factors, developed by SpringWorks Therapeutics. It is designed to block the interaction between YAP/TAZ and TEAD proteins, thereby inhibiting TEAD-dependent transcription that drives cell proliferation and tumor progression in cancers with Hippo pathway mutations. SW-682 binds to the palmitoylation pocket of all TEAD isoforms and has demonstrated downregulation of YAP/TAZ target genes, inhibition of proliferation in vitro, and tumor regression in vivo in preclinical models. The drug is currently being evaluated in Phase 1 clinical trials for advanced solid tumors—including mesothelioma, head and neck cancer, non-small cell lung cancer—and specifically targets patients with or without Hippo pathway alterations who have progressed after standard therapies[1][3][4][5][6][8].
