SW140154 is a small molecule lead compound identified by researchers at the University of Texas Southwestern Medical Center as part of the NCI Cancer Treatment Discovery and Development Network (CTD2N). It was discovered through high-throughput screening for agents that selectively inhibit the growth of lung cancer cells while sparing normal lung epithelial cells. SW140154 has demonstrated efficacy against subsets of both non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC) cell lines. Its growth inhibition profile in SCLC is distinct from conventional chemotherapies like etoposide and topotecan. Mechanistically, the compound induces G1 cell cycle arrest. Research indicates that SARM1 gene expression levels anti-correlate with SW140154 IC50 values, suggesting that SARM1 may serve as a predictive biomarker for drug sensitivity.
