SWAP-70 short hairpin RNA is an RNA interference (RNAi) based therapeutic agent designed to silence the expression of the Switch-associated protein 70 (SWAP-70). SWAP-70 is a multifunctional signal transduction molecule and guanine nucleotide exchange factor (GEF) primarily expressed in B cells, where it regulates critical processes such as polarization, directed migration, and adhesion to endothelial cells. In the context of autoimmune diseases like Multiple Sclerosis (MS), SWAP-70 is implicated in the pathogenic activation and homing of B cells to the central nervous system. By utilizing shRNA to knock down SWAP-70 levels, this therapy aims to modulate B-cell-mediated immune responses and reduce neuroinflammation. Preclinical research using the experimental autoimmune encephalomyelitis (EAE) model has demonstrated that acquired inhibition of SWAP-70 can alter immune cell subsets and ameliorate disease progression.
