Overview

SWI019 is a protein-based, anti-CD19 adapter antibody developed as the soluble “switch” component of Calibr’s switchable CAR-T (sCAR-T) platform, used in combination with the autologous CAR-T product CLBR001 to direct and control T‑cell activity against CD19-positive B cells in cancer and autoimmune diseases.[1][4][5][7] By binding CD19 on B cells and the engineered sCAR on CLBR001 T cells, SWI019 functions as a titratable adapter that turns the CAR-T cells on and off in a dose-dependent manner, enabling potent CD19-targeted cytotoxicity while allowing rapid modulation of activity and mitigation of cytokine release syndrome and neurotoxicity via withholding or reducing SWI019 dosing.[1][3][5][9] The molecule was initially developed by the California Institute for Biomedical Research (Calibr, now part of the Calibr-Skaggs Institute for Innovative Medicines at Scripps Research) and is being evaluated in phase 1 trials in relapsed/refractory B‑cell malignancies and in multiple severe autoimmune diseases as part of the CLBR001 + SWI019 combination.[1][4][5][6][7][8][10]
