Overview

SXR1096 is a first-in-class, potent and selective small molecule inhibitor of the serine proteases kallikrein 5, 7, and 14 (KLK5, KLK7, KLK14). It is being developed as a topical cream for the treatment of Netherton syndrome—a rare and severe genetic skin disorder characterized by overactivity of these proteases due to deficiency in the endogenous inhibitor LEKTI. By inhibiting these targets directly in the skin’s outermost layer (stratum corneum), SXR1096 aims to address both inflammation and barrier dysfunction central to disease pathology. The drug is currently in phase I/II clinical trials in Europe and has received rare pediatric disease designation from the FDA. Sixera Pharma is developing this investigational therapy with potential to become the first approved treatment for Netherton syndrome[1][3][4][5][7].
