Overview

SY-307 is an investigational engineered cell therapy consisting of cord blood-derived natural killer (NK) cells that are genetically modified to express a high-affinity T cell receptor (TCR) targeting the tumor-associated antigen PRAME (PReferentially expressed Antigen in MElanoma), along with interleukin 15 (IL-15). This "off-the-shelf" therapy is designed for immuno-oncology applications, specifically to target and kill cancer cells expressing PRAME. The product leverages the immunogenicity and restricted tissue expression of PRAME, which is found at high levels in various hematologic malignancies such as acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS), as well as multiple solid tumors including melanoma, sarcoma, ovarian, endometrial, lung, and breast cancers. The mechanism involves both direct cytotoxicity from NK cells and targeted recognition via the engineered TCR[1][3][4][5].
