SYD1875 is an investigational antibody-drug conjugate (ADC) composed of a humanized immunoglobulin G1 (IgG1) monoclonal antibody targeting the oncofetal antigen 5T4. The antibody is site-specifically conjugated to a duocarmycin-based linker-drug, specifically valine-citrulline-seco-DUocarmycin-hydroxyBenzamide-Azaindole (vc-seco-DUBA). This design enables targeted delivery of the cytotoxic payload to 5T4-expressing tumor cells. Upon binding to 5T4 on cancer cells, SYD1875 is internalized and releases its duocarmycin payload intracellularly, leading to DNA alkylation and cell death. The drug was developed by Synthon and Byondis for the treatment of locally advanced or metastatic solid tumors expressing 5T4[1][3][4][7]. Phase I clinical trials have been completed in Europe for patients with late-stage or metastatic solid tumors[3][7].
