SYH2072 is a potent and highly selective small molecule inhibitor of aldosterone synthase (CYP11B2), developed by Shanghai Yishi Pharmaceutical Technology (StoneWise) for the treatment of uncontrolled hypertension. By targeting CYP11B2, the enzyme responsible for the final stages of aldosterone production in the adrenal cortex, SYH2072 effectively lowers plasma aldosterone levels. This mechanism addresses the underlying pathophysiology of mineralocorticoid excess, which contributes to resistant and uncontrolled hypertension, as well as potential end-organ damage in the heart and kidneys. SYH2072 is designed to be highly selective for CYP11B2 over the closely related enzyme 11β-hydroxylase (CYP11B1) to avoid interfering with cortisol synthesis, potentially offering a superior safety profile compared to non-selective inhibitors. It is currently being evaluated in Phase 2 clinical trials.
