Overview

SYHA1402 is a highly selective small molecule aldose reductase inhibitor developed for the treatment of diabetic peripheral neuropathy and related complications. Aldose reductase is a key enzyme in the polyol pathway of glucose metabolism; under hyperglycemic conditions, its activation leads to excess conversion of glucose to sorbitol, resulting in cellular and organ injury. By inhibiting aldose reductase (AKR1B1), SYHA1402 reduces sorbitol accumulation and helps preserve nerve function. Preclinical studies demonstrated that it significantly inhibited nerve conduction velocity deceleration and increased Na/K ATPase activity in sciatic nerves. Clinical trials have shown that single oral doses are well tolerated with favorable safety profiles[1][4][5]. The drug is being developed by CSPC ZhongQi Pharmaceutical Technology for indications including diabetic peripheral neuropathy, polyneuropathies, diabetic cardiomyopathies, and type 2 diabetes mellitus[3][2].
