Sym004 is a first-in-class, investigational anti-EGFR therapy composed of a 1:1 mixture of two recombinant chimeric monoclonal antibodies (mAb992 and mAb1024) that bind to non-overlapping epitopes on the extracellular domain III of the epidermal growth factor receptor (EGFR). This dual binding induces rapid internalization and degradation of EGFR, leading to potent inhibition of both ligand-dependent and ligand-independent EGFR signaling. Sym004 has demonstrated superior antitumor activity compared to other anti-EGFR antibodies such as cetuximab or panitumumab in preclinical models, including efficacy in tumors with acquired resistance to these agents. The combination with carboplatin (a platinum-based DNA-damaging chemotherapeutic) and paclitaxel (a microtubule-stabilizing agent) is under clinical investigation for enhanced anticancer effects in solid tumors[1][2][3][4][5].
