Overview

Sym005 is a recombinant monoclonal antibody mixture developed by Symphogen that targets the human epidermal growth factor receptor 2 (HER2). Generated using Symphogen's proprietary mSymplex technology, Sym005 consists of a mixture of systematically selected, non-overlapping monoclonal antibodies cloned from an immunized repertoire. By binding to multiple distinct epitopes on the extracellular domain of HER2, Sym005 is designed to trigger receptor internalization and degradation, block downstream signaling, and engage immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Preclinical studies demonstrated that Sym005 has superior anti-tumor activity compared to trastuzumab and pertuzumab in HER2-positive breast and gastric cancer models, including trastuzumab-resistant cell lines. However, the program was not advanced into clinical development.
