Sym013 is a recombinant pan-HER antibody mixture composed of six humanized immunoglobulin G1 (IgG1) monoclonal antibodies that simultaneously target three members of the human epidermal growth factor receptor family—EGFR (HER1), HER2, and HER3. Each pair of antibodies binds to non-overlapping epitopes on each receptor, resulting in comprehensive inhibition and downregulation of all three targets. This approach is designed to overcome tumor resistance mechanisms that arise from compensatory upregulation within the HER family when only one receptor is inhibited. Sym013 has demonstrated broad antitumor activity in preclinical models across various epithelial cancers—including colorectal, pancreatic, breast, lung, and gastric cancer—and retains efficacy even in models resistant to EGFR- or HER2-targeted therapies such as cetuximab or trastuzumab. In clinical studies for advanced epithelial malignancies, Sym013 showed evidence of disease stabilization and partial response in some patients but was discontinued during phase 1 trials for business reasons[1][2][3][4][5][6].
