Sym015 is a mixture of two humanized immunoglobulin G1 (IgG1) monoclonal antibodies, Hu9006 and Hu9338, that target non-overlapping epitopes in the extracellular domain of the human hepatocyte growth factor receptor (MET; HGFR; c-Met). This dual-antibody approach allows simultaneous binding to the SEMA-a domain of MET, inducing receptor internalization and degradation. By blocking ligand (HGF) binding and inhibiting ligand-induced MET activation, Sym015 suppresses tumor cell growth and proliferation in vitro and tumor growth in vivo. It also induces antibody-dependent cellular cytotoxicity (ADCC). The drug is being developed primarily for solid tumors with alterations or amplification of the MET proto-oncogene—including non-small cell lung cancer (NSCLC), gastric cancer, metastatic colorectal cancer (mCRC), and renal cell carcinoma—with a current clinical focus on NSCLC patients with MET exon 14 skipping mutations or MET amplification. Developed by Symphogen, preclinical studies have shown that Sym015 is well tolerated with an excellent safety profile[1][2][4][5].
