Overview

Sym021 is a humanized monoclonal antibody that targets programmed cell death protein 1 (PD-1). It functions as an immune checkpoint inhibitor by binding to PD-1 and blocking its interaction with the inhibitory ligands PD-L1 and PD-L2. This blockade releases the inhibition of T-cell activity mediated by PD-1 signaling, thereby restoring immune function and promoting T-cell-mediated anti-tumor responses. Sym021 is notable for being derived from a chicken antibody platform, which allows it to bind a unique epitope on PD-1 distinct from those targeted by other clinically approved anti-PD-1 antibodies such as pembrolizumab and nivolumab. The drug has demonstrated high affinity for human PD-1 and cross-reactivity with cynomolgus monkey and mouse PD-1 in preclinical studies. Developed initially by Symphogen and now under development in collaboration with Servier, Sym021 is being investigated primarily for advanced solid tumors in early-phase clinical trials[4][5][6][9].
