Sym022 is a recombinant, human Fc-inert monoclonal antibody that targets lymphocyte activation gene 3 protein (LAG-3; also known as CD223). By binding to LAG-3 and blocking its interaction with MHC class II molecules, Sym022 acts as an immune checkpoint inhibitor. This blockade enhances T cell proliferation and cytokine production, thereby promoting antitumor immune responses. The drug is being developed primarily for the treatment of advanced solid tumors and lymphomas that are refractory to available therapies or lack standard treatment options. Preclinical models suggest improved antitumor effects when used in combination with other checkpoint inhibitors such as anti-PD1 (Sym021) or anti-TIM3 (Sym023). Symphogen originated the drug and it is being co-developed by Servier.
