Overview

Sym024 is a human monoclonal antibody that targets CD73 (also known as 5'-nucleotidase or NT5E), an ectoenzyme involved in the production of immunosuppressive adenosine in the tumor microenvironment. By inhibiting CD73, Sym024 aims to reduce adenosine-mediated immune suppression and enhance antitumor immune responses. The drug is being developed primarily for advanced solid tumors and non-small cell lung cancer. It has been studied both as monotherapy and in combination with Sym021 (an anti-PD-1 antibody). Preclinical data suggest that combining CD73 inhibition with PD-1 blockade may synergistically reverse immune suppression and promote T cell activation. The developer is Symphogen (a subsidiary of Servier)[1][3][4][5][8].
