SYM025 is a human monoclonal antibody designed to target the V-domain Ig suppressor of T cell activation (VISTA), an immune checkpoint protein. VISTA is primarily expressed on tumor-infiltrating lymphocytes and myeloid cells, where it functions as a negative regulator of T-cell activation, thereby contributing to an immunosuppressive tumor microenvironment. By binding to and inhibiting VISTA, SYM025 aims to restore and enhance anti-tumor immune responses. Developed by Symphogen, a subsidiary of Servier, SYM025 is currently being investigated in clinical trials, often as part of combination regimens with other checkpoint inhibitors like the PD-1 antagonist cemiplimab, for the treatment of advanced malignancies including non-small cell lung cancer (NSCLC).
