SYN-114 is an orally bioavailable, potent and selective small-molecule antagonist of the serotonin 5-HT6 receptor that was developed for central nervous system indications, particularly Alzheimer’s disease and other cognitive disorders. It was originally developed by F. Hoffmann-La Roche and later advanced by Synosia Therapeutics (subsequently combined with Biotie Therapies) as a cognitive enhancer based on the role of 5-HT6 receptor blockade in modulating cholinergic, dopaminergic, glutamatergic, adrenergic and GABAergic neurotransmission, which are implicated in neuropsychiatric and neurodegenerative diseases.[1][3][5][7][11] Clinical development progressed through Phase 1 single and multiple ascending dose studies, but the program was ultimately discontinued.[1][3]
