SYN-2869 is a novel, orally active triazole antifungal agent developed by Naeja Pharmaceutical. It belongs to a series of 2(2,4-difluorophenyl)-3-(4-substituted piperazin-1-yl)-1-(1,2,4-triazol-1-yl) butanol derivatives. The compound exerts its antifungal activity by inhibiting fungal lanosterol 14-alpha demethylase (CYP51A1), an enzyme essential for ergosterol biosynthesis in fungal cell membranes. Preclinical studies demonstrated that SYN-2869 is rapidly absorbed with a bioavailability exceeding 45% in mice and shows significant distribution into lung tissue, making it particularly effective in respiratory tract infection models. It was primarily evaluated for the treatment of invasive pulmonary aspergillosis and Candida infections, but development has been discontinued at the preclinical stage.
