Synapsinae (SNG-12) is a small molecule glycine transporter 1 (GlyT-1) inhibitor developed by SyneuRx International (Taiwan) Corp. for the treatment of major depressive disorder (MDD) and suicidal ideation. The drug's mechanism of action involves blocking the reuptake of glycine, an obligatory co-agonist of the N-methyl-D-aspartate (NMDA) receptor, thereby increasing synaptic glycine levels and indirectly enhancing NMDA receptor signaling. This approach aims to address glutamatergic dysfunction associated with depressive symptoms and suicidality. As of late 2022, Synapsinae was in Phase 3 clinical development for MDD and suicidal ideation, and in Phase 2 for psychotic disorders and dementia.
