Overview

SynB1-ELP-(KLAKLAK)2 (also known as SynB1-ELP-KLAK) is a genetically engineered, thermally targeted proapoptotic fusion polypeptide designed for cancer therapy. Developed by researchers at the University of Mississippi Medical Center, the construct consists of three functional domains: an N-terminal cell-penetrating peptide (SynB1) derived from porcine leukocyte protegrins to facilitate cellular internalization; a central thermally responsive elastin-like polypeptide (ELP) biopolymer that remains soluble at physiological temperature but aggregates in response to mild hyperthermia (40–42 °C) to enable localized tumor targeting; and a C-terminal proapoptotic peptide, (KLAKLAK)2 (KLAK), which selectively disrupts mitochondrial membranes to induce apoptosis. In preclinical studies, the polypeptide demonstrated enhanced cytotoxicity and apoptosis induction in breast cancer cells (such as MCF-7) when combined with local hyperthermia.
