Overview

SynB1 ELP2-DOXO is a research-stage biopolymer-drug conjugate designed as a thermally non-responsive control variant of the thermoresponsive conjugate SynB1 ELP1-DOXO. It is composed of three functional components: a cell-penetrating peptide (SynB1) to promote cellular and tumor uptake; an elastin-like polypeptide (ELP2) carrier; and the (6-maleimidocaproyl) hydrazone derivative of doxorubicin (DOXO-EMCH) as the active chemotherapeutic payload, linked via a pH-sensitive hydrazone bond. Under physiological conditions, the ELP2 carrier remains soluble and does not undergo phase transition (which occurs only above 60°C), making it effectively non-thermoresponsive. It has been evaluated in preclinical academic research, specifically in MDA-MB-231 triple-negative human breast cancer xenograft mouse models, to validate the thermal-targeting mechanism of its thermoresponsive counterpart.
