SYNB1344 (SYN-MSUD) is an orally administered, non-pathogenic, engineered strain of *Escherichia coli* Nissle 1917 developed as a "Synthetic Biotic" for the treatment of Maple Syrup Urine Disease (MSUD). MSUD is a rare metabolic disorder characterized by the body's inability to process branched-chain amino acids (BCAAs), specifically leucine, isoleucine, and valine, leading to toxic accumulation. SYNB1344 is genetically modified to express a metabolic pathway that actively consumes these BCAAs within the gastrointestinal tract, thereby reducing their systemic levels and preventing neurological damage. Developed by Synlogic, the program aimed to provide a convenient alternative or adjunct to strict dietary protein restriction. However, development was halted following Synlogic's decision to cease operations and liquidate the company in early 2024.
