Overview

SYNB1891 + atezolizumab is a combination therapy investigated for advanced/metastatic cancers, consisting of SYNB1891, a live, engineered strain of *Escherichia coli* Nissle 1917, and atezolizumab, a monoclonal antibody targeting PD-L1. SYNB1891 is designed to produce cyclic dinucleotides in hypoxic tumor environments, activating the STimulator of INterferon Genes (STING) pathway in phagocytic antigen-presenting cells, thereby stimulating innate immune responses and type I interferon signaling. Atezolizumab acts by inhibiting the PD-L1 pathway, counteracting tumor-mediated immune suppression and facilitating T-cell activity. Together, these agents are intended to synergistically enhance antitumor immune activity via innate and adaptive mechanisms. The combination was evaluated in a phase I trial for safety, tolerability, pharmacodynamics, and preliminary efficacy in patients with advanced/metastatic cancers[1][2][3][4].
