Drug intelligence / Profile preview
SYNB2081 (SYN-GOUT) is an orally administered, non-systemically absorbed Synthetic Biotic developed by Synlogic in partnership with Ginkgo Bioworks for the treatment of gout. It is a genetically engineered strain of the probiotic bacterium *Escherichia coli* Nissle 1917 designed to express bacterial and fungal urate oxidase enzymes and overexpress a urate transporter. Within the gastrointestinal tract, SYNB2081 imports and metabolizes uric acid into 5-hydroxyisourate, which spontaneously degrades into the highly soluble and easily excreted metabolite allantoin. This localized consumption of uric acid in the gut leverages the enterorecirculation loop to lower systemic uric acid levels, offering a potential therapeutic option for gout, particularly in patients with compromised kidney function. Development of SYNB2081 was discontinued in February 2024 when Synlogic ceased operations following the failure of its lead Phase 3 program.
