Overview

Synchotrope TA2M is a DNA plasmid vaccine encoding epitopes of the tyrosinase protein, a key enzyme in melanin synthesis and a well-characterized melanoma-associated antigen. Developed by SuperGen, the vaccine was designed to stimulate a systemic T-cell-mediated immune response against melanoma cells. The "Synchotrope" designation refers to the specific delivery platform, which utilizes a portable pump to provide a continuous, low-dose infusion of the DNA plasmid directly into a lymph node (intranodal administration). This method is intended to maximize the transfection of local antigen-presenting cells, such as dendritic cells, to enhance the presentation of tyrosinase antigens to the immune system. Clinical Phase I studies in patients with Stage IV melanoma demonstrated that the vaccine was well-tolerated and capable of inducing specific immune responses (e.g., to the tyrosinase 207-216 epitope), although objective clinical tumor responses were not observed in early trials.
