SynKIR-310 is a chimeric antigen receptor (CAR) T-cell therapy developed using Verismo Therapeutics' proprietary KIR-CAR platform. It targets CD19, a protein commonly expressed on the surface of B-cell malignancies, using the DS191 binder. Unlike traditional CAR T therapies, SynKIR-310 incorporates killer immunoglobulin-like receptor (KIR) and DAP12 split signaling domains to enhance T-cell activation and co-stimulation, which may prolong antitumor T-cell function and persistence. This design aims to address challenges such as early relapse in patients with advanced B cell lymphomas. The drug is currently being evaluated in phase 1 clinical trials for relapsed/refractory B-cell non-Hodgkin lymphoma (B-NHL), including subtypes such as diffuse large B-cell lymphoma, follicular lymphoma, mantle cell lymphoma, marginal zone lymphoma, mediastinal large B-cell lymphoma, high-grade B-cell lymphomas with MYC/BCL2/BCL6 rearrangements, indolent/aggressive forms of NHL, Epstein-Barr virus positive lymphomas, and others[1][3][4][5][7][8].
