SynKIR-410 is an investigational CAR-T cell therapy developed by Verismo Therapeutics. It utilizes a novel KIR-CAR (Killer Immunoglobulin-like Receptor Chimeric Antigen Receptor) platform, which is inspired by natural killer (NK) cell biology. The therapy is designed to enhance T cell persistence and function in the challenging microenvironment of solid tumors, such as glioblastoma. Unlike traditional CAR-T therapies that rely on the CD3-ζ signaling pathway, SynKIR-410 employs a parallel DAP12 activation pathway for T cells, providing both activation and co-stimulation signals independently of CD3-ζ. This mechanism aims to prolong T cell efficacy and prevent exhaustion during repeated tumor encounters. Preclinical studies have shown sustained anti-tumor activity in models resistant to conventional CAR-T approaches[1][4]. The drug was primarily developed for glioblastoma but its development has been discontinued[1][2][3].
