Overview

SynKIR-420 is an investigational Chimeric Antigen Receptor (CAR) T-cell therapy currently in preclinical development by HLB Life Science and Verismo Therapeutics. It is built upon the proprietary KIR-CAR platform, which utilizes the signaling architecture of natural killer (NK) cell receptors, specifically Killer-cell Immunoglobulin-like Receptors (KIR), rather than the traditional CD3-zeta signaling domain found in first-generation CAR-T therapies. By mimicking the natural multi-chain activation pathways of NK cells, the KIR-CAR design is intended to improve the persistence, metabolic fitness, and anti-tumor activity of T cells within the immunosuppressive microenvironment of solid tumors. While the specific tumor-associated antigen targeted by SynKIR-420 is currently undisclosed, the therapy is part of a pipeline focused on addressing challenging solid malignancies that have historically shown poor responses to conventional CAR-T treatments.
