Synsorb Pk is a synthetic analog of the Shiga toxin (Stx) receptor globotriaosylceramide (Gb3), designed to act as an oral adsorbent agent for the treatment of infections caused by Shiga toxin-producing Escherichia coli (STEC). It consists of diatomaceous silicon dioxide covalently linked to a unique oligosaccharide chain that mimics the natural Gb3 receptor. The drug irreversibly binds and neutralizes Shiga-like toxins in the gastrointestinal tract with the aim of preventing their systemic absorption and subsequent complications such as hemolytic uremic syndrome (HUS). Despite promising preclinical data showing its ability to bind Stx1 and Stx2 and protect cells from cytotoxicity in vitro, clinical trials failed to demonstrate efficacy in reducing mortality or severe complications in pediatric patients with diarrhea-associated HUS. Development was ultimately discontinued after phase 3 trials[1][5][6][7][8].
