Overview

Synstatin E is a functionalized auristatin E microtubule inhibitor developed by Synaffix (now a Lonza company) as part of its toxSYN platform. It is designed as a cytotoxic payload for the construction of antibody-drug conjugates (ADCs). As a derivative of auristatin E, it functions by binding to tubulin and inhibiting microtubule polymerization, which leads to G2/M phase cell cycle arrest and subsequent apoptosis in rapidly dividing cancer cells. Synstatin E is typically used in conjunction with Synaffix's GlycoConnect and HydraSpace technologies to create site-specific, stable ADCs with an optimized therapeutic index. The technology has been licensed to several biopharmaceutical companies, including Elevation Oncology, for use in their clinical-stage ADC pipelines.
