Overview

SYNT-101 is an investigational, once-daily oral treatment for obesity developed by Syntis Bio. It is designed to mimic the metabolic effects of gastric bypass surgery by transiently blocking nutrient absorption in the duodenum (the upper part of the small intestine) and redirecting nutrients to the distal small intestine. This process stimulates natural secretion of satiety and metabolism-regulating hormones, including GLP-1 and PYY, supporting improved glycemic control, weight loss, and energy balance. The mechanism—duodenal nutrient exclusion—leverages a proprietary synthetic tissue-lining (SYNT) technology that delivers a safe, temporary polydopamine coating to catalase-rich tissues like the duodenum; this coating lasts up to 24 hours before being naturally cleared from the body. Preclinical studies have shown consistent weight loss with preservation of lean muscle mass and strong safety/tolerability profiles in both animal models and first-in-human pilot studies[1][3][4][6][8].
