Overview

Syntana-4 is a first-in-class cell-penetrating peptide fusion protein designed to inhibit the Notch signaling pathway at the transcriptional level. Developed by Anastasis Biotec, the drug consists of the cell-penetrating domain of Antennapedia (ANTP) fused via a linker to a truncated, dominant-negative peptide from Mastermind-like (MAML) protein (specifically dnMAML(13-74)). By translocating into the cell nucleus, Syntana-4 prevents the assembly of the functional Notch transcriptional complex, thereby suppressing Notch activity and inducing apoptosis in Notch-driven cancer cells. It has demonstrated preclinical efficacy and tolerability in orthotopic models of breast cancer and is being investigated for the treatment of Notch-mutated cancers, including breast and prostate cancers.
