Overview

Syntansine is a proprietary linker-payload component developed by Synaffix (now a Lonza company) for the construction of antibody-drug conjugates (ADCs). It is a key element of the toxSYN platform and features a maytansinoid payload, specifically Ahx-maytansine, which acts as a potent microtubule inhibitor. The payload is conjugated via a cleavable valine-citrulline (vc) linker and a PABC (p-aminobenzyloxycarbonyl) group, incorporating Synaffix's HydraSpace polar spacer technology. This technology is designed to enhance the stability, solubility, and therapeutic index of ADCs. Syntansine is typically attached to antibodies using GlycoConnect, a site-specific glycan conjugation method that ensures a highly uniform drug-to-antibody ratio (DAR). Preclinical evaluations of ADCs utilizing Syntansine, such as a trastuzumab-based conjugate, have demonstrated superior efficacy and safety profiles compared to conventional ADCs like Kadcyla.
