Syntenin shRNA is a gene-silencing therapeutic candidate designed to knock down the expression of syntenin (also known as Syndecan-binding protein 1 or SDCBP). Syntenin is a PDZ-domain containing protein that regulates the expression and trafficking of various transmembrane proteins, including CD138 and CD86. In the context of multiple myeloma, syntenin expression correlates with poor prognosis, including lower overall survival and shorter response duration. Research conducted at Emory University demonstrated that silencing syntenin using lentiviral-delivered shRNA induces apoptosis in myeloma cell lines (such as RPMI 8226 and MM1.s) and reduces the surface expression of CD86, a protein necessary for myeloma cell survival and drug resistance. This suggests that syntenin is a potential therapeutic target for the treatment of multiple myeloma.
