Synthetic Protectin DX (PDX) analog is a preclinical small molecule being developed by Trienix Pharma as a resolution pharmacology agent. It acts as an agonist for the G protein-coupled receptor 37 (GPR37), mimicking the activity of endogenous Protectin DX, a specialized pro-resolving mediator (SPM) derived from docosahexaenoic acid (DHA). By targeting GPR37, the analog promotes the resolution of inflammation, enhances macrophage phagocytosis of cellular debris and pathogens, and reduces the production of pro-inflammatory cytokines. This mechanism is specifically being investigated for the treatment of severe viral infections, including H1N1 and H5N1 influenza, where it aims to mitigate cytokine storms and prevent or treat acute lung injury (ALI) without the broad immunosuppressive effects typically associated with corticosteroids.
