Drug intelligence / Profile preview
SynuClean-D (SC-D) is a preclinical small molecule inhibitor of alpha-synuclein (α-synuclein) aggregation. Identified through a high-throughput screen of a chemical library, SynuClean-D binds to cavities in mature amyloid fibrils, disrupting their structure, preventing the propagation of protofibrils, and inhibiting the aggregation of wild-type α-synuclein as well as its familial variants (A30P and H50Q). In animal models of Parkinson's disease, such as *Caenorhabditis elegans*, SynuClean-D has demonstrated neuroprotective effects by reducing α-synuclein toxicity and rescuing dopaminergic neurons from degeneration. It is currently developed as a research-use-only compound for the study of Parkinson's disease and other synucleinopathies.
