SYS6011 is a recombinant humanized antibody-drug conjugate (ADC) targeting Nectin-4, developed by CSPC Jushi Biopharmaceutical, a subsidiary of CSPC Pharmaceutical Group. Nectin-4 (Poliovirus receptor-related protein 4) is a cell surface protein that is highly expressed in various solid tumors, including urothelial, breast, lung, and ovarian cancers, while showing limited expression in normal adult tissues. SYS6011 consists of a monoclonal antibody directed against Nectin-4 conjugated to a potent cytotoxic payload via a cleavable linker. The mechanism of action involves the binding of the ADC to Nectin-4 on the tumor cell surface, followed by internalization and lysosomal degradation, which releases the payload to disrupt the microtubule network, leading to cell cycle arrest and apoptosis. SYS6011 is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors.
