Drug intelligence / Profile preview
SYS6043 + ivonescimab is an investigational combination therapy consisting of SYS6043, a B7-H3-targeting antibody-drug conjugate (ADC), and ivonescimab (AK112), a first-in-class bispecific antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor A (VEGF-A). SYS6043, developed by CSPC Megalith Biopharmaceutical, consists of a humanized monoclonal antibody targeting B7-H3 (CD276) conjugated to a topoisomerase I inhibitor payload via a cleavable tetrapeptide linker. Ivonescimab, developed by Akeso Biopharma, is a tetravalent bispecific antibody that simultaneously blocks the PD-1/PD-L1 pathway and VEGF-mediated angiogenesis. In November 2026, CSPC and Akeso entered into a clinical research collaboration to evaluate the combination of SYS6043 and ivonescimab in clinical trials for various solid tumor indications in China, leveraging the synergistic mechanisms of ADC-mediated tumor cell killing, immune checkpoint inhibition, and anti-angiogenesis.
